제목 Dual expression of protein with 2A peptide using In vitro transcription(IVT)
초록 The mRNA technology provides a number of advantages over the conventional delivery of pDNA. The delivery of mRNA offers rapid, robust, and dose responsive expression of target proteins in different types of cells. Co-expression of various proteins can be also accomplished by delivering multiple mRNAs. However, the mRNA technology has a limitation due to its unstable single strand RNA structure and vulnerable to enzyme degration and immune system. To overcome these disadvantages, we have optimized chemically modified mRNA. In this study, we conduct co-transfection with dual expression cassette model. We synthesis mRNA using In vitro transcription. Using 2A peptide, we can get similar quantity of protein between front of 2A peptide cistron and back of 2A peptide cistron.
