제목 Efficient CRISPR/Cas9 delivery as a self-assembled Y-RNA structure for an orthogonal genome editing
초록 The RNA-guided nuclease CRISPR (clustered regularly interspaced short palindromic repeat)/CRISPR-associated (Cas) protein 9 system is a new emerging gene editing approach in the field of gene therapy. However, efficiency of genetic edition of target gene using Cas9 protein and gRNA has not been fully realized. In this study, we prepared the Cas9/gRNA cluster loaded to Y-RNA structure to incorporate multiple sgRNAs for multiplex and simultaneous gene editing. We adjusted the size of gRNA by DNA helper and RNase H and generated Cas9/gRNA cluster. Three distinct Cas9/gRNA complexes can self-assemble to form Y-RNA structure for region-specific (loco-specific) gene editing in mammalian cells. Efficient and programmable gene editing has been demonstrated in MEF cells.
