제목 | Efficient production of cadaverine from galactose in Escherichia coli with reconstructed metabolic pathways |
초록 | Although the biosynthesis of cadaverine has been prospected as an eco-friendly alternative route for polyamides generation, inefficient metabolic ability of microbes due to reduced growth rate and low carbon flux toward the synthesis of desired product made it hard to utilize other biomass-derived sugars. Thus, redesign of microbe is necessary for utilizing those carbon sources with enhanced carbon flux and product formation. In this study, we engineered Escherichia coli to produce cadaverine with rapid assimilation of galactose. Redesign of galactose utilization and cadaverine production pathway with synthetic expression cassettes was performed for maximum activity, while inactivating feedback inhibition and degradation/re-uptake pathways. Finally, the resultant strain, DHK4, produced 8.80 g/L cadaverine with high yield (0.170 g/g) and productivity (0.293 g/L/h) during fed-batch fermentation, which was similar to or better than the previous glucose fermentation. |
