Basic amino acid – conjugated Polyamidoamine (PAMAM) dendrimers enhanced the proton buffering effect and transfection efficiency |
초록 |
Gene therapy has become a promising strategy to treat genetic diseases and cancer. Gene transfer using viral or non-viral vector has advantages and flaws. Viral vector shows high gene delivery efficiency more than non-viral vector. However viral vector tends potent cytotoxicity. We introduced polyamidoamine (PAMAM) dendrimer (generation 4, G4) that is widely used as a non-viral and polycationic vector for gene delivery. We synthesized PAMAM dendrimer (G4)-Histidine-Lysine (PAMAM G4-H-K) / -Ornithine (PAMAM G4-H-O) and compared with the previous report (PAMAM G4–Arginine). Lysine and ornithine are cationic amino acids that contribute to the condensation of DNA and interaction with cell membranes. Histidine has an imidazole ring group that can induce a “proton sponge” effect. We performed the retardation, acid-base titration, cytotoxicity and transfection assay to confirm the characteristics. |
