Analysis of nanoscale intermolecular interactions between polyhexamethylene guanidine and model cell membranes |

A variety of antibiotics, toxicants or antimicrobial compounds interact with the biofilm and require accurate assessment of molecular interactions and bioavailability of existing or newly synthesized molecules. In this study, Surface force apparatus (SFA) measurements and Langmuir trough based tensile measurements are performed to reveal the molecular interaction mechanism with the model membrane to assess the biotoxicity of potential toxic compounds. Polyhexamethylene guanidine (PHMG) was selected as a toxic compound because PHMG contained in the humidifier disinfectant caused many victims in S. Korea due to lack of comprehensive evaluation of PHMG toxicity and interaction mechanism in biofilm. Using SFA and LB tensile measurements, we have shown that only PHMG exhibits strong interactions with biological membrane interfaces and has demonstrated the potential to use this method as a new toxicity testing method. |
