제목 Metabolically Engineered Klebsiella oxytoca for enhanced production of (2R,3R)-butanediol as a raw material for agricultural chemicals
초록 2,3-butanediol exists in three types of stereoisomoers, (2R,3R)-BD, (2S,3S)-BD, and (2R,3S)-BD. Their applications are strongly changed to some special sectors by their different physiochemical properties. (2R,3S)-BD is applicable to a component of cosmetics as an moistening agent. (2R,3R)-BD has an applicability in agricultural industry, which is to activate plants’ own defense systems. Natural Klebsiella oxytoca mainly produces (2R,3S)-BD (> 90% of total 2,3-BD). In order to expand the potential applications of K. oxytoca strain, we engineered the metabolism of K. oxytoca strain by introducing an exogenous gene encoding (2R,3R)-BD dehydrogenase into the site of budC gene in K. oxytoca strain and increasing the expression of of (2R,3R)-BD dehydrogenase further. The final strain could produce up to 106.7 g/L of (2R,3R)-BD [(2R,3S)-BD, 9.3 g/L] in fed-batch fermentation, with a productivity of 3.1 g/L/h and a yield of 0.40 g/g.
