제목 Osteogenic Differntiation of Tonsil-derived Mesenchymal Stem Cells Controlled by Hydroxyapatite in PEG-Polypeptide Thermogel
초록 Understanding the effect of hydroxyapatite on tonsil-derived Mesenchymal Stem Cell (tMSC) differentiation is important for enabling new biomedical applications. We prepared the hydroxyapatite (Hyp) with rod and flower shape which regulated by controlling the concentration of F127. The tMSCs are cultured in the 3D matrixes of in situ formed Poly(ethylene glycol)-poly( L -alanine-co-L-phenyl alanine) thermogel with different shape of Hyp (PEG-PAF/Hyp). The osteogenic differentiation was evaluated by alkaline phosphatease activity assay, immunofluorescence staining, alizarin S staining and mRNA expression of osteogenic gene without osteogenic medium. The results indicated that tMSCs in PEG-PAF/Hyp scaffold promotes osteogenesis. Our PEG-PAF/Hyp scaffold have a good potential for bone regeneration.
