제목 Standardizing the assay procedures of cell-free synthesized cytosol-penetrating antibodies
초록 Antibodies that penetrate and accumulate in cytosol can greatly enrich the options for treating various diseases. Assessment of the cell-penetrating activity of individual antibodies, however, is a time-consuming and complicated process involving the expression and functional assay of purified recombinant antibodies. In this study, we made an attempt to streamline theprocedures for studying cell-penetrating antibodies by integrating the techniques of cell-free protein synthesis and cell-based functional assay. In this new scheme, PCR-amplified genes of cell-penetrating antibodies are rapidly expressed in the reaction mixture for cell-free synthesis, which are then directly transferred to the culture of reporter cells that generate signal in response to the cell-penetrating antibodies. It was required a number of issues to be solved before this scheme was made to work, which will be discussed in this presentation.
