제목 Development of multicistronic vectors containing 2A peptide for IVT mRNA based multiple protein expression
초록 Messenger RNA(mRNA) has drawn much attention as an alternative genetic material that can be expressed target protein instead of pDNA. mRNA provides several advantages over conventional pDNA for the expression of proteins. Previous study we had prepared chemically modified mRNA by in vitro transcription and high protein expression. IVTmRNA has shown considerable effect instead of pDNA and modification increase enzyme stability and reduce innate immune stimulation. When coexpression of more than one gene is required in cells, bi/multicistronic vectors have been used. Conventional multicistronic vectors have been include IRES or other promoters before target gene. These elements are often not expressed protein at the same level. A ‘2A peptide(2A)’ could be good element to replace IRES and other promoters. In this study, we have constructed vector include 2A and prepared IVTmRNA. Transfection and expression of IVTmRNA have been confirmed in various types of cells and cell free system.
