Chitosan is one of the positive charged biocompatible polymer. Chitosan based nanoparticles are formed by ionic gelation of positive charged chitosan and negative charged materials (e.g. Tripolyphosphate, antibody). Amine group of chitosan is reacted with carboxyl group of antibody or PEG-COOH. In this study, chitosan nanoparticles (Chi-TPP NPs) were manufactured by ionic gelation of Chitosan and tripolyphosphate. PEGylated Chi-TPP NPs (PEG/Chi-TPP NPs) were manufactured by EDC/NHS crosslinking of Chi-TPP NPs and PEG-COOH. Antibody conjugated chitosan nanoparticles (PEG/Chi-TPP/Ab NPs) were manufactured by EDC/NHS crosslinking of PEG/Chi-TPP NPs and antibody. The size and zeta-potentials of nanoparticles were measured as under 300 nm and upon +20 mV using Zetasizer Nano, respectively. The shape and aggregation nanoparticles were confirmed by TEM. Chi-TPP NPs were aggregated in pH 7.4 PBS but PEG/Chi-TPP NPs and PEG/Chi-TPP/Ab NPs were uniformly diffused in pH 7.4 PBS. |
