제목 Synthesis and characterization of a bioreducible cationic AB2 miktoarm polymer for efficient gene delivery
초록 Non-viral gene delivery system has received much attention since that may overcome the limitations arise from viral vector. Various cationic polymers and their derivatives, including PEI, have been widely considered as a gene carrier although they induce significant toxicity in biological system. Here, we have synthesized AB2 miktoarm bioreducible and non-reducible cationic polymer, where a hydrophilic poly(ethylene glycol) as A block and a cationic poly(l-lysine) as a B block with or without S-S bond. The cationic miktoarm polymers form nanocomplex with a model gene eGFP-N1, size and zeta were ~150 nm and ±~20 mV respectively. Polyplex composed of the bioreducible miktoarm polymer shows significantly higher transfection and lower cytotoxicity compared to non-reducible studies in MDCK (normal) and KB (cancer) cell lines. In vivo feasible studies also provide evidence of gene expression in liver and kidney whereas shows less hemolytic profile compared to bPEI.
