Tunable gene expression toolbox independent from coding sequence for rational microbial engineering |
초록 |
Customization of protein expression levels is one of the major challenges faced by the field of biotechnology to make synthetic networks reliably display a designed behavior and metabolic networks efficiently produce desired chemicals. Here, we describe a tunable gene expression toolbox that enables producing proteins at various predefined protein levels without being affected by introduced coding sequences. We designed gene structures that can diminish the structural effect of introduced coding sequence on translation and that can show pre-defined protein expression levels. We showed its practical use for developing synthetic circuit and chemical-producing microbial cell factories. Therefore, our toolbox may facilitate the construction of reliable and vulnerable synthetic networks and maximally optimized metabolic networks, and assist the study of protein functions as well.
