제목 Co-amorphization of Linagliptin by Grinding and Ball Milling Methods
초록 Linagliptin is a drug belonging to COX 2 which inhibits DPP-4 enzyme whose oral dosage forms were approved by US FDA in 2011. Linagliptin belongs to BCS Class II due to its extremely low solubility. In this study, grinding and ball milling were employed to manufacture the co- amorphous solid forms between linagliptin and various additives.
Among them, effective were the orcinol and resorcinol as co-amorphization agent. Resulting amorphous materials were pretreated suitably and characterized via PXRD (powder x-ray diffraction) and DSC (differential scanning calorimetry). It was found that the pathway of co-amorphous solid formation was significantly dependent upon the mass ratio between the drug substance and co-amophization agent. As anticipated, the co-amorphous solid powders showed substantially improved solubility compared to pure linagliptin powder. Detailed procedure and experimental data shall be presented in the poster session.
