For the targeted chemotherapy of lung cancer, a single-point mutation of the epidermal growth factor receptor is an important indicator to select an anti-cancer drug in personalized medicine. In this study, the composition of lipids on exosomes were profiled for BEAS-2B, A549, PC9, and PC9GR, to identify specific lipid composition induced by point mutation of EGFR including metastatic T790M. The results showed that the lipid profiles of phosphatidylcholines, sphingomyelins, and amino acids in lung cancer cells were clearly changed compared to those of BEAS-2B cells, and the difference was enhanced according to the mutation level of EGFR. Moreover, the profile of lipids in exosomes showed amplified molecular signals by the metastatic T790M mutantion in PC9GR. This result indicated that lipidomics of exosomes could be used for understanding of exosome-induced metastasis as well as diagnostic biomarker acquired from the informative extracellular vesicles secreted from target cells. |
