Dual delivery of drug and gene using disulfide crosslinked bPEI modified PLGA nanoparticles (NPs) for synergistic therapeutic efficacy |
초록 |
Dual delivery of drugs and gene has been proposed to enhance gene expression or to achieve the synergistic/combined effect of drug and gene therapies. Here we report a multifuctional PLGA hydrophobic core - disulfide crosslinked bPEI cationic shell NPs that can deliver hydrophobic drug and gene simultaneously with potential of targeted delivery. Enhancement in gene delivery efficiency with minimal cytotoxicity by cross-linking of small PEIs via potentially biodegradable linkages was explored. BPEI 600 is thiolated by ring opening reaction with methylthiirane and thiolated bPEI 600 was oxidated in DMSO for 48 hrs. And disulfide crosslinked bPEI coated PLGA NPs were prepared by modified w/o/w double emulsion method. The effective introduction of cationized bPEI was confirmed by various method, and adsorpted pDNA retained luciferase expression activity. |
