For the last few decades, demand of immunoglobulin G (IgG) production has been increased due to its biological importance. To harvest highly purified IgG, affinity chromatography has been commonly used. Conventionally, protein A immobilized agarose gel has been used as a packing material for affinity chromatography, and the target IgG is captured by the immobilized protein A. However, it has some critical limitations which are originated from the intrinsic mechanical properties of agarose gel. Thus, we immobilized protein A to core-shell type polymer supports which have improved mechanical properties, and applied them to IgG purification. |
