It is well recognized that the bioactive molecules such as growth factors and cytokines play an important role for cell adhesion, proliferation and differentiation. Thus, if some constructions with the concentration gradient of bioactive molecules can be prepared, it will become a powerful tool for basic studies, including the determination of optimum growth factor concentration for stem cell differentiation to a specific cell. In our previous study, we fabricated a cylindrical polycaprolactone (PCL) scaffold with gradually increasing pore size and porosity (decreasing surface area) along the longitudinal direction by a centrifugation method. In this study, we fabricated the PCL scaffolds with the concentration gradient of different growth factors via pore surface immobilization of the pore size gradient scaffold with growth factors. The growth factor release behavior of the scaffolds with growth factor gradient were characterized along the longitudinal direction. |
