제목 Bio-electochemical system enabled Acetate Bioconversion to Succinate by mutant Pseudomonas putida
초록 Succinic acid offers great potentials as a platform chemical for synthesis of bio-based plastics, solvents, adhesives, and resins. Currently, succinic acids being produced petrochemically. Succinic acid can be produced by microbial fermentation process using reductive TCA pathway. Hence, in this study, fermentative by-product acetate used as a substrate for the biotechnological production of succinate, using industrial host Pseudomonas putida. During anoxic acetate assimilation, one NADH generate from two acetate after entering glyoxylate shunt and recycling of NADH require additional electron acceptors to maintain redox balance. Maintaining redox balance is crutial for acetate assimilation. for this purpose, a mutant P.putida strain with acetyl-CoA synthetase over-expression and succinate dehydrogenase deletion was applied to the carbon electrode of bioelectro chemical system (BES).
