Injectable Polypeptide Hydrogels with Stimuli-responsiveness for Biomedical Applications |

Genetically engineered polypeptides with stimuli-responsiveness have been studied as advanced biomaterials for biomedical applications because both precisely controlled amino acid sequence and monodisperse molecular weight could provide finely tuned biophysical property as well as environmental responsiveness while immunogenicity and biocompatibility could be optimized at the amino acid sequence. We report herein that injectable polypeptide hydrogels with stimuli-responsiveness were developed by temperature-triggered physical crosslinking of (1) elastin-based polypeptide (EBP)-calmodulin (CalM) fusion proteins and (2) EBP-resilin block copolymers for in vivo applications. Especially, the EBP-CalM fusions showed multiple responsiveness when bound to specific ligands while the the EBP-resilin block copolymers having high resilient property coupled with elasticity. Dynamic hydrogelation in response to multiple stimuli would be potential for functional tissue engineering. |
