Rheumatoid arthritis (RA) is a chronic inflammatory disease that causes pain, stiffness, and inflammation of the synovial membrane in the joints leading to articular destruction. In this work, hyaluronic acid (HA) was grafted with tyramine (Tyr) and used for the preparation of injectable HA-Tyr hydrogels by radical crosslinking with hydrogen peroxide and horse raddish peroxidase. Intra-articular injection of in situ forming HA-Tyr hydrogels encapsulating dexamethasone (DMT) resulted in successful treatment of RA with reduced IL-6 and PGE2 levels in collagen-induced arthritis animal models. Histological analysis with hematoxylin & eosin (H&E) staining also confirmed the therapeutic effect of injectable HA-Tyr hydrogels with DMT. The novel injectable HA-Tyr hydrogel systems will be investigated in detail for further clinical applications. |
