Hydrogenotrophic methanogen(HM) is an attractive microorganism that can produce methane using CO2 and H2. In this study, biological methane production by HM has been investigated in mesophilic condition. The sludge from an anaerobic digester of waste water treatment plant was utilized as a seed culture and supplied in cylinder type reactor (0.2L) that can maintain atmospheric pressure by gas tight piston. In order to enrich HM, the sludge was adapted in mix gas of CO2/H2. The reactor produced methane of 80ml per day. One ml of culture broth from the reactor was injected into 120ml serum bottle containing fresh basal medium of 20ml for serial sub-culture. The enriched culture was produced methane of 19 ml per day. To confirm being of HM, 16SrRNA gene was amplified by specific primers 519F/934R and phylogenetic analysis were conducted using eztaxon-e and MEGA7. As a result, the isolate was similar to Methanobacterium arcticum and performance for methane production was evaluated in-depth. |
