PLGA microsphere/alginate system of dual growth factor delivery for stem cell differentiation |

While the pivotal role of growth factors on stem cell differentiation has been widely known, it is believed that continuous release of growth factors may not be enough to elicit a maximized differentiation potential of stem cells and therefore a temporally and/or spatially controlled release system is definitely required. In this study a dual growth factor delivery system that exhibits a temporal release of individual growth factor is being realized using PLGA microsphere and alginate hydrogel. Once PLGA microspheres are manufactured using W1/O/W2 double emulsion method, they are then subjected to a micro-encapsulation in alginate by applying electric field. With each growth factor enclosed, the efficacy of dual delivery system will be evaluated using stem cells. This study can provide a basic understanding of how stem cells respond to individual growth factors that are delivered in a temporally controlled manner from dual release system. |
