Rapid DNA Amplification on Silicon Based Micro-PCR Chip Controlled by Designed Thermal Cycling System |

Micro-PCR chip and thermal cycling system were developed that permit rapid DNA amplification. The micro-PCR chip was fabricated on silicon wafer through MEMS processing and was bonded with glass wafer. The thermal cycling system uses thermoelectric element and temperature sensor for control the operating temperature. The micro-PCR chip has 5㎕ reaction volume and the thermal cycling system attains maximum thermal gradient of 15℃/sec and steady-state temperature accuracy of ±0.1℃. In results, target DNA of beta-globin was amplified on the micro-PCR chip controlled by the designed thermal cycling system in as rapid as 15 min. In experiments, Bovine Serum Albumin (BSA) that coats the reaction chamber surface accounts for stable amplification. DNA amplification process using this micro-PCR chip and designed thermal cycling system is not only considerably faster than commercial PCR process, but also have an illustrative significance towards developing lab-on-a-chip.
