Gene therapy holds enormous potential for therapeutic intervention of a broad range of genetic diseases, including infectious diseases, gene-related disorders, and cancer. Improving gene transfer efficiency is prerequisite for successful clinical gene therapy. To address the needs, polymeric gene carriers should be carefully designed to overcome several delivery barriers. In this study, we developed multifunctional non-viral vectors tailored to meet specific needs of genetic drugs, such as intracellualr distribution of plasmid DNA or siRNA. The synthetic carriers showed the way of improving tranfection efficiency by mimicking viral infection mechanisms. |
