Bone marrow stromal cells (BMSCs) have the ability of differentiation into the multiple lineages such as osteoblasts, chondrocytes and adipocytes. In this study, a biomimetic ECM network that is derived from target tissue cells is introduced in 3D scaffold. We hypothesize that this self-assembled ECM-based structure may provide stem cells with a microenvironment for cell adhesion and differentiation. A porous composite scaffold was fabricated using a mixture of PLGAHydroxyapatiteβ-tricalcium phosphate . After MC3T3-E1 preosteoblast cells were seeded into the scaffolds and cultured for 1 or 3 weeks and the cell-scaffold constructs were decellularized. Once rabbit BMSCs were seeded into the decellularized scaffolds, the BMSCs-scaffolds constructs were cultured in an osteogenic medium for 4 weeks. Mineralizaiton was confirmed in von Kossa and Alizarin red S stainings. Nude mouse implantation also confirmed the efficacy of decellularized composite scaffold. |
