A Multifunctional Polymeric Scaffold Incorporating Magnesium Hydroxide, Decellularized Extracellular Matrix, and PDRN for Kidney Tissue Regeneration |

Poly(lactide-co-glycolide) (PLGA) is one of the most common polymers that has been used to prepare tissue engineering scaffold. Although previous studies have showed potential of PLGA scaffold, safety issues were raised. To improve the biocompatibility of PLGA scaffold, we constructed scaffold with magnesium hydroxide (MH) and decellularized kidney extracellular matrix (ECM) for kidney tissue regeneration, but it was not sufficient to restore the kidney function. Polydeoxyribonucleotide (PDRN) ameliorated wound healing by reducing inflammation, and angiogenesis. Here, we verified the application of a multifunctional scaffold containing PDRN. PDRN improved the proliferation and wound closure of kidney-derived cells as well as angiogenesis. Moreover, compared to PLGA/MH/ECM scaffold, PLGA/MH/ECM/PDRN scaffold enhanced biocompatibility. Consequently, these results suggest that the multifunctional PLGA scaffold containing PDRN improved regenerative potential for kidney tissue. |
