제목 ECM-mimetic topographic substrates as cell chips for  in vitro cytotoxicity test platforms
초록 Pre-clinical trial including in vitro and in vivo animal testing is essential to find out the cytotoxicity, and it is also important to screen out the candidates in new drug development. In this study, we propose the extracellular matrix (ECM)-mimetic topographic substrates as new pre-clinical trial platforms for investigating anticancer drug-induced cytotoxicity of stem cells. The morphologies and behaviors of stem cells were controlled by the ECM-mimetic topographic substrates, eventually affecting the cells fate (i.e., survival and death) against the common chemotherapeutic agent cisplatin. Most importantly, the cisplatin-induced cytotoxicity was prevented by the appropriate inhibited gap junctions of stem cells on the ECM-mimetic topographic substrates unlike the flat substrates. Our finding demonstrated the ECM-mimetic topography may be a crucial parameter for cytotoxicity test, which can be used as new pre-clinical trial platforms in drug development.
