Biodegradable PLGA nanoparticles (NPs) have been widely applied to carriers for protein delivery due to the controllable release time. Nevertheless, harsh conditions damaging proteins and poor intracellular uptake of PLGA NPs are difficult problems to conquer for efficient protein delivery as usual. The aim of this study is to optimize formulation conditions of protein using PLGA NPs for intracellular uptake and sustained release in cytoplasm. A monoclonal antibody, 3D8 scFv, was used as a model protein to investigate sustained cytoplasmic delivery from PLGA NPs because the targeting site for the antiviral activity is cytoplasm. Physico-chemical properties of PLGA NPs and biological activity of 3D8 scFv were evaluated by various methods. Cellular uptake and cytoplasmic release of PLGA NPs were investigated with confocal laser scanning microscopy (CLSM) and FACS. In addition, further studies plan to be performed for investigating the protecting effect of 3D8 scFv from virus infection. |
