제목 Purification and functional reconstitution of CXCR2 derived from E. coli
초록 Chemokine receptors interact with chemokines, a family of small cytokines. These interactions cause the G protein-mediated intracellular signaling. Certain chemokines and their receptors are associated with cancer and various inflammatory diseases. CXCR2(C-X-C Motif Chemokine Receptor 2) is one of them. CXCR2 is generally overexpressed in most tumor cells and affects to the progression of cancer through cell cycle, metastasis, and angiogenesis. We purified CXCR2 using E. coli expression system and assembled it into a nanodisc with membrane scaffold protein and lipid. Nanodisc is a well-known platform to get the membrane protein in native form in vitro. We also characterize the CXCR2-embedded nanodisc by size and functionality.
