Novel Poly(L-lactic acid)-Poly(ethylene glycol) Multiblock Copolymers as a Material for Cartilage Regeneration |

Hydrophobic/hydrophilic multiblock copolymers, containing poly(L-lactic acid) PLA and poly(ethylene glycol) PEG segments in their main chain, were synthesized through a two-step reaction: triblock copolymers PLA/PEG/PLA were prepared by ring opening polymerization and then were chain extended resulting in mutiblock copolymers. The synthesized mutiblock copolymers were characterized by various analytical techniques such as 1H NMR spectroscopy, gel permeation chromatography, differential scanning calorimetry, contact angle analyzer, and universal testing machine. Multiblock copolymers exhibited higher hydrophilicity and elongation property compared to PLA as a control material. In vitro cell culture studies were conducted on film-type multiblock copolymers using primary rabbit chondrocytes. Less hydrophilic copolymer showed better cell attachment, cell proliferation and cell differentiation among synthesized multiblock copolymers. |
