Chitosan-based hydrogel is a good candidate for cartilage regeneration, since it shares some characteristics with a variety of glucosaminoglycan (GAG) which is one of the major macromolecules found in articular cartilage. In this study, we developed thermosensitive chitosan-Pluronic (CP) hydrogel and investigated in vitro evaluation of the injectable CP hydrogel. In the result of MTS assay, CP hydrogel was more biocompatible than alginate gel, control material. In addition, gene expression, cell number and GAG content of CP hydrogel were significantly increased as compared with those of alginate gel. In protein release test, BMP-7 showed sustained release behavior from CP hydrogel, indicating a good growth factor carrier for cartilage regeneration. Therefore, the obtained results suggest that CP hydrogel is a prospective injectable material for cartilage regeneration as well as a new biomaterial for various biomedical applications. |
