High temperature (30~40°C) decreased the autotrophic astaxanthin production in H. pluvialis. The O2- (superoxide anion radical) burst was the main source of the trouble to slow induction at high temperatures. MV (methyl viologen), artificial O2- generator was used to demonstrate the phenomenon of O2--induced retardation of astaxanthin production under autotrophic conditions. However, the productivity of astaxanthin was inversely improved by Haber-Weiss reaction (Superoxide-Driven Fenton reaction), in which O2- was readily oxidized to O2 by the ferric (Fe3+) with the explosive OH• (hydroxyl radical) generation in cells. Subsequently, Haematococcus cells rapidly accumulate astaxanthin to the cytoplasm to survive in unfavorable conditions by extreme oxidative stress. Consequently, Fe2+-Fe3+ redox cycle amplification is an alternative promising strategy for the effective astaxanthin production under autotrophic culture in H. pluvialis. |
