In this report, we conjugated polyethylenimine-dexamethasone (PEI-Dexa) to the biodegradable backbone polyglutamic acids derivative (PEG-Glu) to form PEG-Glu-PEI-Dexa. PEG facilitates the formation of polyplexes with improved solubility, reduced aggregation, lower cytotoxicity, and possibly decreases opsonization with serum proteins in the bloodstream. The complex formation of PEG-Glu-PEI-Dexa with plasmid DNA was confirmed by agarose gel electrophoresis method. The size distribution and the zeta potential value of PEG-Glu-PEI-Dexa with pDNA were measured using Zeta-potential & Particle size Analyzer ELS-Z. In gel retardation assay, PEG-Glu8-PEI-Dexa and PEG-Glu10-PEI-Dexa were completely retarded at or above weight ratio (polymer/DNA) of 14 and 12, respectively. From the results of the transfection study, PEG-Glu-PEI-Dexa showed much enhanced transfection efficiency and lower toxicity in various cell lines. |
