Fabrication of a label-free and portable LSPR biosensor for the highly sensitive detection of Salmonella in Pork samples |

Here, we developed a label-free portable plasmonic biosensor device to enable ultra-sensitive and selective detection of Salmonella in pork samples. Concretely, a plasmonic sensor utilizing the self-assembly of the gold nanoparticles was utilized to conduct high density deposition on a transparent substrate, which produced longitudinal wavelength extinction shifts through the LSPR signal. The plasmonic aptamers were deposited on AuNPs using a dipping method. The fabricated LSPR sensor chip revealed a highly sensitive and low limit detection of Salmonella of ~104 cfu/mL under the optimal assay conditions. SEM analysis clearly demonstrated the morphologies of bacteria and aptamer binding interaction. Moreover, the aptamer based LSPR biodevices were established and applied to determination of food poisoning caused by Salmonella in actual pork samples. The cost-effective plasmonic chip also enabled highly selective recognition of Salmonella in a multiplexed matrix. Overall, the developed plasmonic portable bio-devices are an innovative, elegant, rapid, ultra-sensitive and multiplex method of identifying bacteria that can be utilized by the agricultural and food industries |
