제목 Replicable Three Arm Junction RNA Nanostructure for Target Specific Gene Silencing
초록    We achieved a large-scale synthesis of RNA nanostructures using rolling circle transcription (RCT). Various RNA nanostructures can be prepared by inserting the siRNA sequences within the each arm of nanostructure. It is important to understand and investigate the clear requirements for RNA nanostructures with gene silencing activity. In this study, we have confirmed and clarified which form of RNA nanostructure is appropriate to induce sequence-specific gene silencing in vitro. Depending on their difference in structure and direction of siRNA, they showed markedly different phenomenon in dicer processing and gene silencing activity. Dicer processing has revealed that blunt Y-RNA structure cannot be processed, while Y-RNA structure with overhangs are easily cleaved in 18 hours. In addition, the gene silencing efficiency is enhanced when 3’-end of sense strand is facing outward in blunt Y-RNA structures and facing opposite direction in Y-RNA structures with overhangs.
