A better understanding of protein and protein interactions is a powerful tool to obtain from multiplex protein detection technologies, and because the efficiency and convenience, encoded bead was usually used for this detection method. However, high-throughput detection of encode bead is still challenging because of the difficulty of binding between labeled secondary protein and target protein. In this study, we report ligand immobilization on polydiacetylene-coated and surface enhanced Raman scattering-encoded beads (PDA-SERS beads) for label-free detection. The fluorescence intensities of biotin-conjugated PDA–SERS beads were increased by the increasing concentration of streptavidin. Also, we could detect streptavidin with detection limit of 2ⅹ10-8 M throughout the fluorescence intensity without additional labeling step. It is expected that the system using the PDA–SERS beads can be useful as a powerful tool to analyze biopsy in the multiplex way. |
