Recently, bone morphogenetic proteins (BMP) are dominantly used for bone treatments. However, the amounts of BMP for treatments is much higher than physiological concentration, because of short half-life and instability of the BMP. In this study, we prepared the MPEG-b-(ε-caprolactone–ran-α-chloro-ε-caprolactone) (MC-Cl), and then chlorides of the MC-Cl are modified to azides. The BMP-2 containing alkyne group was chemically introduced MC-Cl by cycloaddition. BMP-2 linked MC-Cl (MC-BMP2) aqueous solution was mixed with human periodontal ligament stem cells (hPDLSC) and injected to dorsum of nude mouse. The injected solution immediately formed a gel-state. hPDLSC in MC-BMP2 hydrogel were proliferated and differentiated to osteoblast like cells. We observed osteoblastic phenotypes and gene expression through histological methods and qRT-PCR respectively. In conclusion, we confirmed that MC-BMP2 hydrogels could increase a half-life and a stability of the BMP-2. |
