A large-scale production of secondary metabolites in bioindustry is a strenuous process, as over-production of metabolites could be toxic and also cause significant metabolic burden to their hosts. Therefore, the precise control of the metabolite production during fermentation is of great importance. Here, we present the modular design approach to build a high performance inducible promoter system that displays a large dynamic range. To this end, an inducible promoter system was divided into two separate functional modules, promoter and regulatory modules. We first constructed the four different inducible promoter cassettes that combine a strong promoter with four operator-repressor pairs that were known to function in Streptomyces. Then, these modules were individually optimized using the indigoidine-based promoter assay system. We believe that the synthetic inducible promoter system we report here could be a useful tool in modulating secondary metabolite production in Streptomyces. |
