제목 Design of PpIX-conjugated pH-sensitive polymeric nanoparticles for Photodynamic Therapy
초록 An effective tumor-targeted drug delivery system for photodynamic therapy was generated by designing ligand-mediated nanoparticles with stable formulations of a hydrophobic photosensitizer. Novel folic acid (FA)-conjugated amphiphilic block copolymers were synthesized. The 2,4-diacetyl deuteroporphyrin IX dimethyl ether (Pp IX)-conjugated amphiphilic block copolymers containing folic acid ligands were synthesized. Photosensitizer-loaded copolymeric nanoparticles (FA-PEG-P(Asp-Hyd)-Pp IX) were prepared by micelle formation in an aqueous solution. The results of in vitro release tests showed that the release profiles of Pp IX from the nanoparticles were strongly pH-dependent and influenced by the loaded amount of photosensitizer. FA-PEG-P(Asp-Hyd)-Pp IX nanoparticles were not cytotoxic without light and showed good cellular uptake. In addition, FA-PEG-P(Asp-Hyd)-Pp IX nanoparticles showed marked phototoxicity compared with free PpIX.
