Bioreducible Poly(ethylene glycol)-pheophorbide a conjugate for the preparation of self-quenchable nanoparticles with controllable phototoxicity |

This study is aimed to design and synthesize a poly(ethylene glycol)-pheophorbide a conjugate with bioreducible disulfide linkers between PEG and each PhA moiety (PEG-(ss-PhA)2) for controllable photodynamic therapy (PDT). The amphiphilic PEG-(ss-PhA)2 conjugate formed self-assembled nanoparticles with self-quenching properties in aqueous media. It was confirmed that PEG-(ss-PhA)2 nanoparticles are photoinactive with no fluorescence and singlet oxygen generation under light treatment in aqueous condition. The nanoparticular structures could be dissociated by reductive cleavage of the disulfide linkers, followed by an efficient dequenching process. Based on present results, we propose that PEG-(ss-PhA)2 nanoparticles could potentially be used in PDT with minimal unfavorable phototoxicity. |
