제목 Tissue Engineering of small caliber vascular grafts
초록 A tissue engineering approach has been developed to produce arbitrary lengths of vascular graft material from smooth muscle and endothelial cells that were derived from a biopsy of vascular tissue. Recently, many studies have been reported that mechanical stimuli enhanced the development and function of engineered tissues. So it is likely to be necessary that the development of elastic scaffolds can maintain their mechanical integrity and deliver the mechanical signals to adherent cells during mechanical strain application.
In this study, PLGA and poly(L-lactide-co-caprolactone)(PLCL) copolymer were synthesized. There polymers were characterized by 1H-NMR, GPC, DSC and so on. Scaffolds were easily fabricated by using solvent-casting and particle-leaching technique, and each structure of those was characterized by SEM. As a result of tensile test, PLGA scaffold was broken at 20 percent strain, but PLCL scaffold was elongated by 500 percent. Vascular grafts(I.D. 0.5cm) were fabricatred from novel PLCL and sequentially seeded with rabbit vascular smooth muscle cells(SMCs) and endothelial cells. After 5 days static culture, the grafts(n=4) were grown in vitro in a pulse duplicator system(bioreactor) for 7, 14, 21, and 28 days. Analyses of tissue included Histology, SEM, and pharmacological reactivity of engineered vessels. This study demonstrates the feasibility of tissue engineering of viable, surgically implantable small caliber vascular grafts and the important effect of a 'biomimetic' in vitro environment on tissue maturation and extracellular matrix formation.
