Efficient recovery of fatty acid from the recombinant strain, namely E. coli MG1655 disrupted cells is extremely important in the biosynthesis of Biofuel from renewable sources. Flocculation of E. coli high-pressure homogenized (02 passes, 5000 psi) cell suspensions with different flocculants were applied to improve the process of centrifugation and extraction due to very small size of the cell debris (i.e., nanoparticles) [1-2]. Four different structures and charge properties synthetic polymers Chitosan, Polyethyleneimine, CTAB and PDADM have been used as the flocculants. Here we demonstrate that the best result was CTAB at a concentration 640 ppm. In addition, hexane/isopropyl alcohol and ethyl acetate/ethanol systems showed relatively good extraction power and less toxic. Under the optimum conditions, the total lipid recovery from recombinant E. coli cell was 67%. |
