제목 Inhibition of smooth muscle cell proliferation using curcumin-loaded PLGA nanoparticles
초록 Abstract
Restenosis occurs in approximately 30-40% of patients following percutaneous transluminal coronary angioplasty (PTCA) for treatment of atherosclerosis. Proliferation of the smooth muscle cells from the arterial wall into the lumen of the artery is considered as a major cause of restenosis.1 In order to inhibit vascular smooth muscle cell proliferation, drug must be delivered at a high concentration for a prolonged period of time.2,3 Therefore, local sustained drug delivery systems have been under investigation to treat the regional pathophysiologic process of restenosis. 4-6
In this study, we evaluate the feasibility of local delivery of curcumin, an anti-proliferative drug of smooth muscle cells, using nanoparticles formulated with a biodegradable PLGA (poly (DL-lactide-co-glycolide)) as sustained-release system for the prevention of restenosis. Drug containing PLGA nanoparticles were prepared by the emulsification method. Variable factors such as the type and concentrations of stabilizer, sonication out-put, polymer concentrations, could be the important factors for the formation of PLGA nanoparticles. The release of the drug was sustained over 4 weeks in vitro, and the curcumin loaded PLGA nanoparticles showed an antiproliferative effect against smooth muscle cells comparable to free curcumin.
