Biodegradable nanogels for photodynamic therapy (PDT) |

Photodynamic therapy (PDT) is a form of phototherapy using nontoxicity photosensitizers (PSs) which make reactive oxygen species (ROS) killing cells when they are exposed a light source with appropriate wavelength and oxygen molecules. Although PDT has potential possibility for treatment of tumor and other diseases, it has many difficulties such as low solubility and low selectivity for the target site. Herein we describe synthesis of biodegradable nanogel as PSs carrier for stable dispersion of PSs and increasing selectivity to tumor. Chlorin e6 (Ce6) was used as PSs and encapsulated in nanogel via sonication. Synthesized nanogel was characterized by DLS, AFM and GPC and the encapsulation of Ce6 was characterized by UV-Vis spectroscopy, photoluminescent spectroscopy and fluorescent microscopy. Cell uptake and phototoxicity of Ce6-loaded nanogels are confirmed by UV-Vis spectroscopy, fluorescent microscopy and EZ-Cytox cell viability assay kits after 3-hr incubation with HeLa cell. |
