In the culturing the cell in the sheet, the rapid detachment of cell from the sheet is important for stabilizing the cell at the low temperature, which occurred the phase-separation of sheet polymer. To achieve the rapid swelling/deswelling rate, the sheet material should be porous due to the larger surface area and consist of chemically modified molecular structure. Poly(N-isopropylacrylamid) (PNIPAAm) is known for temperature-sensitive substrates for the sheet culture. From the reasons, nano-fiber using the electrospining was prepared for the sheet culture with larger surface area, which facilitated the rapid cell sheet detachment. In addition, comb-type grafted poly(N-isopropylacrylamid) (PNIPAAm) was synthesized and processed into the nano-fiber. In the experimental, the morphology and surface area of the substrates were investigated using SEM and BET sorption. The swelling/deswelling rate was measured by dynamic vapor sorption. The cell detachment rate was evaluated using the smooth muscle cells. From the results, the electrospining is promising technique for accelerating the cell detachment in the sheet culture. |
