Kidney diseases occur in a numerous of ways like glomerular disease, diabetes, hypertensive nephrosclerosis, denovascular and other vascular diseases. Even, the decellularization and recellularization techniques has been established the organ like kidney, due to its complex structure, still has limitations for organ transplants and has several effects after surgery. In this paper, an attempt is made to fabricate artificial kidney scaffold using decellularization system. Rat glomerulus endothelial cells in acellular scaffold is cultured and the response of endothelial cell is observed. Rat kidneys were extracted from 12 weeks old rats and cannulated for the decellularization. Kidneys were decellularized successfully with 1% SDS and 1% triton X-100. The seeded endothelial cells were well attached and grow on the vessel walls in ECM kidney scaffold at 37℃ and 5% of CO2 humid conditions for 3 days and 1week. Distribution of endothelial cells and lining in recellularized kidney were observed through histological analysis. In addition, we observed the formation of the tubular structure of the blood vessels in the kidney. Further histological experiments and molecular analysis are also carried out. |
