제목 Oral Controlled Release of Nifedipine using Squeeze Osmotic Tablet System
초록
The osmotic pump runs by directly using osmotic pressure difference as its energy source. The osmotic pump tablet system for oral administration has many advantages such as constant delivery rate and thereby reduced risk of adverse reactions, extended action period and hence improved patient compliance, and in vivo predictability of release rate in the basis of in vitro data. Nifedipine (ND) gastrointestinal therapeutic system (GITS) affords rate-controlled release and once-daily administration. ND is practically insoluble in water with solubility less than 10 g/ml, a well known and most widely used coronary vasodilat or from the group of dehydropyrine derivatives.
The aims of study were to prepare squeeze osmotic pump tablet system (SQZ) by novel mould device. The SQZ was composed of one or more cylindrical squeeze-push layer (squeeze-disc, Sd) and centered drug layer. The SQZ has been found to be able to deliver nifedipine at a controlled rate up to one day. Release patterns were affected by molecular weight of polymers, amount of polymers and excipients. We observe the release pattern and the membrane morphology of novel tablet by HPLC and SEM, respectively.
