Hematopoietic stem cells (HSCs) are continuously stimulated by chemical and physical interactions with bone marrow or umbilical cord niches. Interactions between HSCs and their niche play an important role in proliferation and differentiation of HSCs. To clarify the mechanism of the interaction between HSCs and their niche, many studies have been focused on properties of niches, for instance, substrate elasticity, charge and so on. In this study, we investigated the proliferation, multipotency and erythropoietic differentiation of HSC on tropoelastin and the hydrogels with positive charge (chitosan, poly-l-lysine) and negative charge (alginate), respectively. We found that poly-l-lysine significantly increased the cell number in expansion step without changing expression of surface markers related to their stemness. Thus, poly-l-lysine would be a useful substrate to enhance the proliferation and differentiation of HSCs for clinical-grade generation of red blood cells. |
