제목 Gene delivery system for targeting the artery wall using low molecular water-soluble chitosan
초록 Gene therapy is a method for the treatment of diseases with introducing the gene-engineered materials into a patient with gene-deficiency disease or cancer to produce a therapeutic protein in a patient’s cells. The various gene carriers, chitosan, a natural cationic polysaccharide, is continued to attract a great deal of interest as a candidate non-viral vector for gene delivery because of its low cytotoxicity and high positive charges. Targeted gene carrier was synthesized to target artery wall cells using LMWSC. AWBP-PEG-g-LMWSC was synthesized by conjugaion the artery wall binding peptide (AWBP) to the end of PEGylation LMWSC as a gene carrier to target artery wall cells. Characters of the synthesized AWBP-PEG-g-LMWSC were analyzed by measuring FT-IR, 1H-NMR, zeta-potentiometer and atomic force microscopy (AFM). The formation of plasmid/LMWSC complex was proved and transfection efficiency in 293T cells was evaluated. The cytotoxicity of LMWSC also was determined by MTT assay.
