Encapsulation of enzyme –fluorophore conjugates within poly(ethylene glycol) microparticles for detection of organophosphorus compounds |

Biosensors based on acetylcholinesterase(AChE) inhibition have been developed and used for environmental monitoring. In this study, we fabricated poly(ethylene glycol) microparticles containing the conjugate of pH-sensitive fluorophore (SNAFL-1) and AChE for the detection of organophosphorus compounds. AChE conjugated to a SNAFL-1 were encapsulated in PEG microparticles by suspension photopolymerization and the entrapment of conjugates were verified with Conforcal Laser Scanning Microscope. The pH-sensitive dye of SNAFL-1 has a dual emission peaks which allow the pH sensitivity of the fluorophore to be correlated to the ratio of the fluorescent intensities at the two peaks according to the environmental pH changes. By conjugating AChE with SNAFL-1, changes in the microenvironmental pH due to the enzyme reaction between AChE and the analyte of interest were used to fluorescently detect low concentration of the analyte. These PEG microparticles containing enzyme-fluorophore conjugates can be applied for PEBBLE(Probe Encapsulated By Biologically Localized Embedding)sensors and Micro-total Analytical System. |
